Recently, I tried to follow the steps described in  "User instructions and tutorials of the synergyfinder package", Authors: Liye He, Krister Wennerberg, Tero Aittokallio1, and Jing Tang, using data file called "mathews_screening_data" as an example. I succeeded to obtain the results at each step described in the paper mentioned above with this data file.

My question is the following: How to load my own input data file and to follow the same steps : input data, input data visualization, drug synergy scoring, visualization of sinergy scores.

I tried to do this, but the message error is that my file is not found !

You can use csv or excel to have all the necessary columns and then use read.table or read.csv functions to load the file into R. Then you can follow the instructions.
