selection of interesting genes for a KS test in TopGO

KS test does not require an arbitrary p-value threshold, so why to I need to provide a threshold in a function to select genes of interest? I do not understand, what is that 'geneSel= topDiffGenes' for?

The geneSel is the selection of genes from the universe which are the genes provided. Otherwise the topGOdata doesn't know which is the gene set of interest.
