Sample expression calculation in rna seq

Hello....!! My supeevisor asked me to calculate total expression of sample in rna seq. He want to get the total expression (i.e sum of expression of all genes in a single sample). Then he want to compare the expression of samples. For example sum of all genes in sample1 vs sum of all genes in sample 2. Is it logical? And if it is can you recommend me a method to calculate this. And also let me know about a paper for reference. Thanks.

The support site is for technical help with the packages. Consider asking at biostars.org and consider to discuss with your project group first what is logical or not. Figuring out which analysis to do depending on your project is nothing the internet communities can help with. A sum of a count matrix per gene is done by colSums.
