Hello,

I downloaded CRISPRseek a while ago and find it to be a wonderfully useful tool! I primarily utilize it to find predicted off-target effects of created gRNAs (just to provide some context for the question).

When looking at the Off-target results output files, I find two quantifiable prediction measures: Score and gRNA efficacy. I thought that I had a proper understanding of these terms, but a brief look at how each are calculated showed me that I was incorrect...

I now understand that score does take into account mismatch position in its calculation and that it is not just a generalized horology score. I also understand that gRNA efficacy is purely calculated by the gRNA alone as opposed to its interaction with genomic DNA (previously refereed to as gRNA efficiency, which seems to shed some light on its interpretability).

But I think that I need a specific definition of the both of them to really nail the difference in my brain, as I could not find any explanation in the vignette or a solid definition of gRNA efficacy online.

More specifically: how would I interpret the meaning of a score of 0 and a score of 100 in the score column. How would I interpret the meaning of a score of 0 and a score of 1 in the gRNA efficacy column

Thank you very much!
